Trigonelline (TG): Cognitive Benefits, Memory Enhancement, and Molecular Effects in Mice

Trigonelline (TG), a plant alkaloid present in coffee,

as well as in fenugreek seeds (plant and condiment) and radish, thus offers cognitive benefits. The study reveals its effects on memory and spatial learning, from both a cognitive and molecular point of view, in mice.

The study: Experiments carried out in animals reveal that:

oral administration of TG to mice for 30 days provides significant improvement in spatial learning and memory, as assessed by the water maze test; signaling pathways related to nervous system development, mitochondrial function, ATP synthesis, inflammation, autophagy and neurotransmitter release are significantly modulated in the intervention group, as observed by whole-genome transcriptomic analysis of the hippocampus; TG suppresses neuroinflammation by downregulating proinflammatory signaling factor-mediated activation; the levels of inflammatory cytokines TNF-α and IL-6 are significantly decreased, as revealed by quantitative protein analysis; levels of neurotransmitters dopamine, norepinephrine and serotonin are significantly increased in the hippocampus.

Taken together, these results, which will need to be validated in humans, suggest the effectiveness of TG in the prevention and reduction of age-related cognitive decline and the benefit of supplementation.
