The 31-year-old Ontarian shares the lead with Belgium’s Thomas Detry, who shot 67. They total 134, minus 10.
A shot behind is Sepp Straka (66).
A Charlotte resident, Hughes has completed each of the last three seasons in the top 15 of the PGA for play on the greens.
The 29-year-old Detry is a PGA rookie, but he has a solid track record on the European Tour.
He had three putts of 20 feet or more.
Austrian Straka won the Honda Classic in February, earning a first PGA victory. He birdied five of his last seven holes.
Mark Hubbard (69), Garrick Higgo (66) and Scott Stallings (67) are each two strokes behind.
Mississippi golfer and co-leader following 18 holes, Davis Riley brought back a card of 71. He slipped to seventh place at seven under, with Nick Hardy (67) and Stephen Jaeger (68).
Nick Taylor (70 for 138), Adam Svensson (70 for 139), Michael Gligic (68 for 140) and Adam Hadwin (69 for 142) are the other golfers representing Canada.
