“Discovering MPI 8: A Revolutionary Anticoagulant Compound with Low Risk of Bleeding”

[메디컬투데이=최재백 기자] A new anticoagulant compound has been discovered that does not pose a significant risk of bleeding.

The results of a study that revealed a new anticoagulant compound MPI 8 that does not have a high risk of bleeding were published in ‘Nature Communications’.

When a blood clot formed in the body breaks apart, it blocks blood vessels and prevents blood flow to major organs such as the brain, heart, and lungs, resulting in stroke, pulmonary embolism, and heart attack.

Anticoagulants are used prophylactically in people with a high risk of thrombosis or patients with atrial fibrillation due to their tendency to coagulate, or to prevent thrombosis in patients who have undergone heart valve, knee, or pelvic surgery.

Currently used anticoagulants act on the entire coagulation mechanism involved in thrombus formation and excessively inhibit coagulation, which can rather cause bleeding.

Experts mentioned that taking anticoagulants can make it difficult to control even small wounds or bleeding. They explained that it was only in the last 20 years that novel direct anticoagulants (NDCs) had been developed that might reliably inhibit blood clotting while reducing the risk of severe bleeding.

A research team from the University of British Columbia and the University of Michigan found that MPI 8 might reduce the risk of unwanted bleeding while inhibiting clot formation by inhibiting just one of the molecules involved in blood clotting.

The research team set polyphosphate as the main treatment target, explaining that polyphosphate is a polyanionic molecule with negative charges in several regions.

Then, they mentioned that MPI 8 is an abbreviation of ‘Macromolecular Polyanion Inhibitor 8’, and MPI 8 has a very low cation density, so it normally moves freely in the body’s bloodstream, and when it binds to polyphosphoric acid, the target substance, it becomes charged. It is explained that the density can be increased to be able to bind specifically.

The research team expressed this characteristic of MPI 8 as tunability.

They now confirmed that MPI 8 effectively prevented clot formation in mice without toxicity and bleeding risk, and hoped to evaluate its efficacy in larger animals and even humans in future studies.
