Haaretz reports that a woman and a man in their 70s who were critically injured died in hospital after the attack.
Two others are injured – an elderly man with serious injuries and a younger man with moderate injuries, according to the Magen David Adom ambulance service.
The alleged perpetrator was shot and killed by a police officer. The police have identified the man as a Palestinian from Salfit in the occupied West Bank.
The attack happened in the middle of rush hour traffic on Sunday morning in Holon, which is located between Tel Aviv and Ben Gurion International Airport.
Magen David Adom says the victims were found in three different places 500 meters apart in the city. The perpetrator attacked people both in a park and at a gas station.
The police say that large police forces have been deployed, and that a search is being carried out with a helicopter and other resources in the area in case more people are involved.
Tensions have risen sharply after two top leaders of Hezbollah and Hamas were killed in attacks in Beirut and Tehran blamed on Israel, and there are growing fears of full-scale war in the area, in addition to the war in Gaza.
Earlier on Sunday, at least four people were killed in an Israeli airstrike against a tent camp for displaced Palestinians at al-Aqsa hospital in Gaza. Thousands of people live in the tent camp after their houses were destroyed in the war.
