Could a “simple” cocktail of dietary supplements help improve the cognitive decline inherent in Alzheimer’s disease ? An international research team (Sweden, Great Britain and Turkey) has just completed a study1 of phase II which suggests that the answer is… yes!

The cocktail in question is composed of nicotinamide ribosideof carnitineof serine and of N-acetylcysteine (at 1 g, 3.73 g, 12.35 g and 2.55 g per dose, respectively). It is inspired by a formulation already marketed by the company ChromaDex under the name of Tru Niagen®.

The nicotinamide riboside is a derivative of vitamin B3 and a precursor of NAD+, notably involved in the production of ATP and the synthesis of neurotransmitters. Carnitine and serine are two amino acids (the basis of proteins), while N-acetylcysteine is a synthetic derivative of another amino acid, cysteine. All these micronutrients target in particular the stimulation of mitochondria, the energy suppliers of our cells. According to the most recent studies, mitochondrial function is altered in people suffering from Alzheimer’s and Parkinson’s. Brain cells would be more exposed to oxidative stress, less well supplied with energy and, ultimately, more prone to degeneration and death.

The participants in the study received a daily dose of the cocktail for 28 days, then two daily doses until the end of the trial, the total duration of which was 84 days. The researchers measured a 29% improvement in cognitive function (compared to 14% in the placebo group) as well as better liver and kidney health.

This trial on humans, the results of which need to be confirmed in a larger phase III study, confirms previous studies on Alzheimer’s and Parkinson’s diseases on mouse models which had already shown that the same cocktail optimized brain and liver metabolism .

Note that in two previous studies, a similar cocktail had significantly improved the liver markers of patients with fatty liver2 and favored faster recovery of patients who contracted the Covid-193.
